Barrie has developed a reputation as a defenseman with a nose for the net. The leading scorer among defenseman in each of his four full junior seasons with Kelowna of the Western Hockey League, Barrie was selected by the Colorado Avalanche in the third round (No. 64) of the 2009 NHL Draft.

The son of former NHL player and Tampa Bay Lightning co-owner Len Barrie, Tyson Barrie led the Avalanche as a rookie with an average ice time of 21:35 per game. After scoring his first NHL point with an assist in a 4-3 shootout win over the Minnesota Wild on Feb. 14, 2013, he had his first multipoint game in his next game, two days later. On Feb. 18, 2013, Barrie scored his first NHL goal in a 6-5 win against the Nashville Predators.

In a 1-0 victory against the Predators on March 30, 2013, Barrie became the first rookie defenseman in Avalanche history to score an overtime winner.

In the 2013-14 season, Barrie's 13 goals were the most by an Avalanche defenseman since John-Michael Liles scored 14 in 2006-07. Barrie also had three overtime winners among his five game-winning goals.

A native of Victoria, British Columbia, Barrie had two assists against Minnesota in his NHL Stanley Cup Playoff debut on April 17, 2014, including one on Paul Stastny's overtime goal that gave Colorado a 1-0 lead in its first-round series. But after the Avalanche took a 2-0 lead in the series, Barrie played only 7:14 in Game 3 before a knee-on-knee hit knocked him out for the rest of the playoffs. Colorado lost Game 3 and, eventually, the series in seven games.

A healthy Barrie returned in 2014-15, when his 53 points made him the fourth Colorado defenseman to score 50 in a season. He bumped his offensive numbers up to 14 goals and 57 points in 2017-18, then finished with 14 goals and 59 points in 2018-19, when the Avalanche advanced to the second round of the playoffs.

Barrie's time in Colorado came to an end on July 1, 2019, when the Avalanche traded him to the Toronto Maple Leafs. He signed a one-year contract with the Edmonton Oilers on Oct. 10, 2020 and led NHL defensemen that season with 48 points. Barrie played three seasons for the Oilers until he was traded to the Nashville Predators for defenseman Mattias Ekholm on Feb. 28, 2023.

He signed a one-year contract with the Calgary Flames on Oct. 3, 2024.
