Hughes, 19, completed his second NCAA season with the University of Michigan in the Big Ten Conference. The defenseman led the team in scoring with 33 points (five goals, 28 assists) in 32 games after having 29 points (five goals, 24 assists) in 37 games as a freshman last season.
He will travel to Vancouver on Tuesday.
"Quinn is an exceptional skater, strong on the puck and plays with creativity and vision," Canucks general manager Jim Benning said of Hughes, who Vancouver selected with the No. 7 pick in the 2018 NHL Draft. "He was a leader on his team at the University of Michigan and has had success at the international level. We're excited for Quinn to join our team and continue to grow his game as a member of the Vancouver Canucks."
Hughes (5-foot-10, 170 pounds) played for the United States in the 2018 and 2019 IIHF World Junior Championships, winning a silver medal this year.
